Interleukin 4 (IL-4), encoded by the IL4 gene, is a pleiotropic cytokine known primarily for its role in type 2 immunity. IL-4 is mainly produced by mast cells, Th2 cells, eosinophils and basophils. By binding to its receptor IL-4R, this cytokine exerts many biological effects, including stimulation of proliferation of activated B and T cells, and differentiation of B cells into plasma cells. IL4 is considered a canonical regulatory cytokine because alternative activation of myeloid cells was first observed after stimulation with Th2 lymphocyte-derived IL4. IL4 is structurally and functionally related to another immunomodulatory cytokine, IL13. IL-4 has been found to be increased in breast, prostate, lung, renal cell, and other types of cancer, and IL-4R is overexpressed in many types of cancer, suggesting that IL-4 has a significant impact on tumor progression. This product is designed as a tool for the delivery and expression of TF-tagged human IL-4 mRNA for research. The product leverages the lipid nanoparticle (LNP) technology platform for simple and efficient delivery of hIL4-TF mRNA to a variety of mammalian cells in vitro and in vivo. The LNPs used are formulated with SM-102, DSPC, cholesterol and DMG-PEG2000 at an optimal molar concentration for a high rate of encapsulation and efficient mRNA delivery. The hIL4-TF protein is about 19 kD, consisting of hIL4 and a C-terminal TF tag (15 amino acids). The GenPept accession number for hIL4 (153 amino acids) is NP_000580.

Application & Handing
Upon receiving product, briefly pulse spin before opening to ensure product is at bottom of container. It is important not to spin for too long as this may rupture mRNA-LNPs. Do not vortex. Work with mRNA-LNPs on ice and minimize the time that the product spends at room temperature. After handling the product during experiments, return immediately to ice. mRNA-LNP products should only be handled with certified RNase-free reagents and consumables. Use of filtered pipette tips is highly recommended.
