ICOS ligand (ICOSL), also known as cluster of differentiation 275 or CD275, is a glycosylated transmembrane protein belonging to the B7 family, encoded by the ICOSLG gene. ICOSL is expressed on the surface of antigen-presenting cells and non-hematopoietic cells under inflammatory conditions. By binding to inducible T cell costimulator (ICOS), ICOSL triggers distinct intracellular signaling cascades and delivers costimulatory signals that promote T cell activation and differentiation. The ICOS/ICOSLG signaling pathway has been shown to promote antitumor T cell responses as it is activated in Th1 and other Teff cells, or promotes tumor responses when triggered in Tregs cells. Both agonistic and antagonistic monoclonal antibodies targeting this pathway may be used in cancer immunotherapy. This product is designed as a tool for the delivery and expression of TF-tagged human ICOSL mRNA for research. The product leverages the lipid nanoparticle (LNP) technology platform for simple and efficient delivery of hICOSL-TF mRNA to a variety of mammalian cells in vitro and in vivo. The LNPs used are formulated with SM-102, DSPC, cholesterol and DMG-PEG2000 at an optimal molar concentration for a high rate of encapsulation and efficient mRNA delivery. The hICOSL-TF protein is about 36 kD, consisting of hICOSL and a C-terminal TF tag (15 amino acids). The GenPept accession number for hICOSL (302 amino acids) is NP_001350699.

Application & Handing
Upon receiving product, briefly pulse spin before opening to ensure product is at bottom of container. It is important not to spin for too long as this may rupture mRNA-LNPs. Do not vortex. Work with mRNA-LNPs on ice and minimize the time that the product spends at room temperature. After handling the product during experiments, return immediately to ice. mRNA-LNP products should only be handled with certified RNase-free reagents and consumables. Use of filtered pipette tips is highly recommended.
