Chemokine CXCL9 is a small cytokine that belongs to the CXC (C-X-C motif) chemokine family. It is encoded by the CXCL9 gene and has the functions of inducing chemotaxis, promoting differentiation and proliferation of leukocytes, and causing tissue extravasation. CXCL9 regulates immune cell migration, differentiation and activation through the CXCR3 receptor. The CXCL9/CXCR3 axis affects the polarization of tumor-associated macrophages (TAMs) that exert regulatory activity in the tumor microenvironment. Studies have shown that CXCL9 and its two family members, CXCL10 and CXCL11, promote tumor suppressor lymphocyte infiltration in solid tumors through CXCR3. Furthermore, the action of CXCR3 has been shown to be critical for successful immune checkpoint inhibition in preclinical cancer models due to the recruitment and activation of T cells. The CXCL9 gene expression has identified as the most powerful marker for predicting immune checkpoint blockade response in multiple tumor types. It is very meaningful to study the relationship between the CXCL9/CXCR3 axis and the immune checkpoint inhibitor PDL-1/PD-1. It has been reported that blocking the PDL-1/PD-1 axis in T cells may trigger a positive feedback loop at the tumor site via the CXCL9/CXCR3 axis.
This product is designed as a tool for the delivery and expression of CXCL9-TF mRNA for research. The product leverages the lipid nanoparticle (LNP) technology platform for simple and efficient delivery of CXCL9-TF mRNA to a variety of mammalian cells in vitro and in vivo. The LNPs used are formulated with SM-102, DSPC, cholesterol and DMG-PEG2000 at an optimal molar concentration for a high rate of encapsulation and efficient mRNA delivery. The CXCL9-TF fusion protein is approximately 15 kD, consisting of full-length CXCL9 (125 amino acids) and a C-terminal TF tag (15 amino acids). Application & Handing
Upon receiving product, briefly pulse spin before opening to ensure product is at bottom of container. It is important not to spin for too long as this may rupture mRNA-LNPs. Do not vortex. Work with mRNA-LNPs on ice and minimize the time that the product spends at room temperature. After handling the product during experiments, return immediately to ice. mRNA-LNP products should only be handled with certified RNase-free reagents and consumables. Use of filtered pipette tips is highly recommended.
